Porphyrin Metabolism

چکیده
The suggestion of Duesberg (1) that prolonged ingestion of allylisopropylacetylcarbamide (Sedormid)’ may have induced acute porphyria in a human led later workers to produce this disorder in rabbits (2) and rats (3) by oral administration of the drug. Although this experimental procedure has facilitated the study of acute porphyria, the possibility remains that some of the porphyrin excreted in this metabolic disturbance originates from the action of intestinal bacteria (4). This possibility has now been excluded by use of the embryonated hen’s egg in which injection of Sedormid into the yolk sac, followed by examination of the allantoic fluid for porphyrins, provides a situation analogous to oral administration of the drug to animals with subsequent examination of the urine and feces for porphyrins. Such experiments have demonstrated that the accumulation of tissue and excretory porphyrins in Sedormid-treated embryos is comparable to that occurring in acute porphyria. In addition, congenital deformities and retarded growth were noted.

substances, the main one of which he called haematoporphyrin (porphurouspurple). Three years later Schultz published the first case of porphyria. In 1911 Glinther classified the diseases of porphyrin metabolism from published cases, including his own. Since then, numerous papers have been published and various classifications put forward. The latest classification is that from Brodie et al,2 in...
متن کاملUrinary porphyrin excretion in hepatitis C infection.
A high prevalence of hepatitis C virus infection in porphyria cutanea tarda in some populations suggests a close link between viral hepatitis and alteration of porphyrin metabolism. Moreover, there is evidence of a role of porphyrinopathies in hepatocarcinogenesis. The aim of our study was to obtain data on the prevalence and patterns of heme metabolism alterations in patients with chronic hepa...

The clinical history, laboratory, and necropsy findings in a patient with terminal jaundice and disordered porphyrin metabolism are presented. A retrospective diagnosis of erythropoietic protoporphyria is considered.

ECENT STUDIES” 2, 3 have emphasized that in human beings, photosensitivity occurs as a result of two fundamentally different disturbances of porphyrin metabolism. In porphyria cutanea tarda, in which the excessive porphyrin formation is believed to take place in the liver, symptoms are seldom manifest before adult life.’ Its porphyria erythropoietica (congeisital, photosensitive), where large a...

A 42 year old heterosexual male with symptomatic human immunodeficiency virus infection presented with a 2-week history of tense blistering skin lesions following azidothymidine therapy. Urinary porphyrin excretion confirmed the diagnosis of porphyria cutanea tarda. The blisters resolved following the withdrawal of the drug but recurred when rechallenged. Three other cases of porphyria cutanea ...
